The output file is still without subtitles when you use A S S files.
So the missing tag on the header does not cause the problem.

Tried 3 hours to get the subtitles shown but couldn't find any solution.

I used nearly the same settings as you except for my Encoding which was on Auto, not UTF-8, and language was set to Eng (but that doesn't affect the hardcoding process anyway) and I had no problems hardcoding an A.S.S sub file onto an MP4. And it didn't even have the zlib muxing mode property either so it's likely only PGS subs need it.
I'd say it seems that it might not be a problem with the header but something else. What settings are you using under "Video"?
In theory, theory and practice are the same. In practice, they are not.

Your settings look fine but I'm not personally able to test CUDA encoding as I have AMD graphics cards currently. I used x264 as my encoder in my successful test run (slower but can produce better quality/efficiency).
So yeah, try using x264 instead. Keep same settings for everything else. And use "Default render" for subtitle mode.
In theory, theory and practice are the same. In practice, they are not.

Okay, so I used the settings you prefered but still no subtitles.
This problem is really difficult to solve... :(

I also extracted the A.S.S file from the MKV and "muxed out" these subs via mkvmerge.
Afterwards I imported the MKV file without subs in MediaCoder played preview (as expected, no subs shown)
Added the subs, played preview, subs showed up so I rendered it but finally no subs in the output file.

Man, this is so frustrating!

I also tried another program called VidCoder.
With that program I can "burn in" the subtitles and it worked very well.
But it's very very slow and the UI is not as innovative as MediaCoder...

For me as a software developer this really sounds like a massive bug!
I hope the service guys here know this bug and will fix this with the next update..

Maybe you can try it out with your settings.
Here is my MKV File packed with WinRar.

It's the character encoding. Playing back the extracted sub and the video in SubtitleEdit shows that none of the character encoding styles available in MediaCoder can interpret it which is why it can't hardsub them.
Seems that MediaCoder has quite a few issues with non-English languages. I've found that files that have Korean characters in their name or filepath can't even be converted...
In theory, theory and practice are the same. In practice, they are not.

Thanks for your answer but I think you're wrong.

I tried it with an english subbed movie, too.
Without umlauts or special characters...
Exact the same behavior like before.

I guess the character encoding isn't the reason for this misbehavior!

I'm using VideoConverterUltimate now and it works perfectly.
Maybe I go back on MediaCoder if a new version is available

Did you report this bug or shall I do it?

What version are you using? Because I haven't had a lot of issues with english subs in my testing runs (I personally do not hardsub my videos since softsub support is pretty common nowadays). Also, it's possible for English subs to use something other than the MediaCoder supported character encodings so it's best to definitely check what the subs are encoded in. Download the program "SubtitleEdit" and open the sub file, it'll show what encoding it's using up in the top toolbar.
I haven't submitted a bug report. Considering I'm able to at least get English subs to work, it's not really a viable to consider it a bug right now.
In theory, theory and practice are the same. In practice, they are not.

I had the same problem about burning in the sub using the AviSynth, and I solve it using the Haali for DirectShowSource (in my case, when I updated the K-lite Codec Pack, I set this configuration). And it kept all the animations, fonts, colors from the original play-back.
